Wild-type p53 protein has been shown to stimulate BER in extracts of cells. In terms of BER activity, lower levels of beta-pol through MMS treatment did not hinder activity as much as the protein was affected. Other studies have suggested APE and p53 function as a complex (Offer et al., 2001), yet our data do not support this hypothesis.

Sensitivity of p53 cells to MMS, suggest that some cancers may be preferentially sensitive to chemotherapeutic base-damaging agents similar to MMS produce damage that is repaired by BER. Studies, along with ours, indicate that p53 is playing a role in BER pathways. Expanding our understanding of p53 involvement in DNA repair seems like a logical step to producing better therapeutic regimens for treatments of various cancers. (Abstract shortened by UMI.)
