Stroke has emerged as a major public health concern, and ranks third as the leading cause of death or permanent disability. Many risk factors have been associated with stroke including diabetes, HTN, and hypercholesterolemia. Currently, it is not known what role dietary cholesterol plays in chronic inflammation and progression of HTN to stroke. The SHRSP gives researchers an excellent experimental model of human essential HTN, insulin resistance, and dyslipidemia. Thus, 4 week weanlings were fed either 0% cholesterol (control), 2%cholesterol (2%chol) or 0.2% oxidized cholesterol (0.2%oxichol) for 30 days (w/w). Blood pressure components were assessed using femoral artery cannulation. Plasma lipids were measured with enzymatic kits. Hepatic C-reactive protein mRNA was measured by RT-PCR. All data was expressed as +/-SE and analyzed individually by repeated measures MANOVA. Statistical significance was defined as p < 0.05. Results. HR differed significantly between cholesterol fed groups (p < 0.05). Whether 2%chol or 0.2%oxichol the same changes in PP were observed p < 0.05). A decrease in HDL was apparent in cholesterol fed animals (p < 0.05). Significant increase in PL was found in 0.2%oxichol treatment (p < 0.05). Oxidized cholesterol feeding significantly increased expression of CRP in the liver tissue (p < 0.05). Conclusion . Oxidized cholesterol feeding in the SHRSP may exacerbate HTN leading to increased stroke potential. These data suggest a relationship with inflammation, as seen in the rise in HR and PP, suggesting possible changes in the vessel wall. A decrease in HDL and increase in PL, suggesting vessel repair, expression of CRP, which additionally is recognized to promote inflammation leading to endothelium dysfunction.
