Parkinson's disease (PD) is a neurodegenerative disease characterized by progressive loss of neurons in areas of the brain responsible for controlling movements. Current pharmacological treatment leaves much to be desired and may even facilitate cell loss, thereby accelerating the rate of decline. Adenosine antagonists, particularly adenosine A2A receptor antagonists, have gained attention as potential therapeutic agents for the treatment of PD owing to their highly localized expression within the striatopallidal pathway and their resistance to tolerance. The present study investigated the roles of A1 and A2A receptor antagonism in reversing pimozide-induced tremulous jaw movements (TJMs), a rodent model of parkinsonian tremor. Results showed significant reductions following non-selective adenosine antagonist treatment with caffeine. Surprisingly, the A1 antagonist, CPT, produced the greatest reductions. Although treatment with the A2A antagonist, SCH 58261, decreased TJMs, results were not significant. Further research is necessary to gain a more comprehensive understanding of A 1 and A2A antagonism as well as the effects of treatment duration on parkinsonian models such as TJMs.
