Novel polymeric structures and drug release mechanisms for cancer therapy. - 127 p.

This dissertation describes the efforts toward the improvement of drug delivery systems with novel polymer designs. Incorporation of polymers can produce water soluble, nontoxic, long circulating drug delivery vehicles that can transport drugs lacking these properties. A challenge facing this field is the design of polymer carriers with high degrees of drug incorporation. In Chapter 2, a new method is described to produce dendrimers capable of loading multiple functional molecules on their periphery. Another problem is the method by which the drug is removed from the polymer. Chapter 3 describes the use of acetals as drug linkages with tunable rates of release. The final chapters of this work describe the design, demonstration, and implementation of micelles sensitive to near infrared light.
