Molecular mapping of important rootstock traits would facilitate grape rootstock breeding efforts through the identification of closely linked, co-segregating molecular markers useful for marker assisted selection. As a primary step towards achieving this goal, an SSR based genetic map was created from an F1 population (cross '9715') derived from rootstocks Ramsey (Vitis champinii), and Riparia Gloire (V. riparia), which was expected to segregate for resistance to root-knot nematodes, salt tolerance, vigor, and sex expression. The map covered all 19 Vitis linkage groups, with a total combined map length of 1304.7 cM, averaging 6.8 cM between markers, and expected genome coverage of over 92%.

This genetic map provided the framework for mapping the root-knot nematode resistance locus, N1, to the end of linkage group 18. This dominant source of resistance is derived from Ramsey, and is strongly linked to two SSR markers and one AFLP marker derived from a bulked-segregant analysis study. The locus controlling sex expression was mapped to linkage group 2, close to an SSR marker.

Genome-wide and chromosome-wide variation in recombination rates was observed between three mapping populations of varying Vitis spp. parentage. Suppressed recombination in distantly related Vitis species was detected, which may hinder future efforts directed at the fine-scale mapping of disease resistance genes.

The 9715 population showed continuous variation for the average root angle per genotype, analyzed from both herbaceous and dormant cuttings. A preliminary QTL study identified five QTLs associated with average root angle, ranging from 9.3% to 14% in their explanation of observed phenotypic variation. Root angle in young cuttings may be associated with the vigor potential of these individuals as rootstocks.

A rapid 18-day screen for tolerance to high saline conditions was developed, and used to assess the salt tolerance of Ramsey and Riparia Gloire. Both individuals showed low levels of leaf percent chloride and foliar salt stress symptoms, suggesting that salt tolerance may not segregate in the 9715 population. Nonetheless, results obtained for other rootstock genotypes tested were similar to those obtained in previous screens using greenhouse and field studies, suggesting that this method is reliable as a fast screen for young vines.
