Ongoing depletion of atmospheric ozone has led to increased levels of solar UV-B radiation (290–320 nm) that reach the earth's surface affecting all biological systems. Plants are particularly vulnerable to changes in UV-B radiation levels since they need sunlight for photosynthesis. The goal of this research was to increase our understanding of the different mechanisms that makeup defenses against UV-B damage in plants. My approach was to determine if new proteins would accumulate in plants in response to UV-exposure. I found that a new 68-kD protein accumulates upon UV-B radiation of Arabidopsis plants and is restricted to UV irradiated tissues. Appearance of this protein was observed in all ecotypes and mutants of Arabidopsis tested. I present evidence that this protein corresponds to a covalent crosslink between large and small subunits of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and that its formation is detectable as early as 2 min after UV-B radiation. These results suggest that accumulation of this crosslinked polypeptide is a direct consequence of UV-damage, which could serve as a good indicator of UV-damage to cellular proteins and may become a useful tool to monitor generalized damage by UV.
