520 $a This work describes the cloning of zebrafish <italic>rho kinase 2</italic> and <italic>myosin regulatory light chain 2a</italic> and <italic>2b</italic>. The involvement of these genes in regulating cell motility in culture together with their expression in gastrula suggested that they might regulate gastrulation movements. Functional analysis shows that reduction of Rok2 function, or expression of a constitutively active Mlc2a disrupts gastrulation. We correlate impaired dorsal movement with disruption of polarized morphology in cells expressing dominant negative Rok2. Cell transplantation reveals that both cell intrinsic properties and intercellular interactions mediate acquisition of polarized cell morphology. Molecular epistasis analysis, and the ability of Wnt5 and Wnt11 to modulate Rok2 intracellular distribution identify Rok2 as a target of a non-canonical Wnt pathway regulating polarized cell behaviors that underlie convergence and extension.
520 $a As gastrulation ends, ventral and dorsal blastoderm cells meet and close over the yolk, forming the tailbud. Despite detailed analysis of cell movements within the tailbud, the genes required for tailbud morphogenesis are not known. Our genetic studies identify synergistic regulation of these cell movements by genes that regulate C&E in gastrula, <italic>pipetail/wnt5, knypek, </italic> and <italic>no tail</italic>. Therefore, tailbud formation involves a continuation of gastrulation mechanisms.
