Herpes Simplex Virus Type 1 (HSV-1) infection is a model for much of what is known concerning viral transcription kinetics, evasion of host defenses, and latent infection. Efficient global analysis of HSV-1 infection using microarray technology has great potential to further research in many aspects of virology. With this goal in mind, a custom DNA microarray with probes for both viral and cellular transcripts was developed and validated. A detailed analysis of HSV-1 infection in different cell types was performed to examine differences in HSV-1 transcription and host cell response dependent on cell type. Wildtype HSV-1 expression was similar in HeLa, B7, and SK-N-SH cells. Data from the cellular probes indicate a cell type-specific immune response in B7 fibroblasts, but not in SK-N-SH cells, supporting observations by others. Additionally, similar infections using a mutant virus, 27<italic>lacZ</italic>, which is incapable of expressing the critical ICP27 regulatory protein, provided evidence of a sub-division of the viral early kinetic class of transcripts.
