Although development and homeostasis of multicellular organisms is critically dependent upon apoptotic cell death, regulation of the cell death process is not fully understood. These studies were designed to identify factors that contribute to Sindbis virus (SV) and antioxidant-induced cell death and to better understand the cell death process following SV infection. The first set of experiments shows that glutamate excitotoxicity contributes to SV-induced death <italic>in vitro</italic> and <italic>in vivo</italic>, that SV-induced death can be either apoptotic or necrotic and that bystander cell death occurs in uninfected cells. The second set of experiments shows that poly (ADP-ribose) polymerase activation contributes to SV-induced apoptotic cell death and examines the role that cyclin-dependent kinase activation plays in cell death following SV infection. The last part of this study identifies cell cycle and cell death regulatory factors involved in antioxidant-induced growth arrest and cell death in colorectal carcinoma cells. These studies contribute to understanding the regulation of apoptotic cell death, a critical process which leads to disease if unregulated.
