A set of 23 consensus chloroplast simple sequence repeat (ccSSR) primer pairs were constructed using the tobacco chloroplast sequences. All primer pairs produced amplicons after PCR employing genomic DNA from members of the Chenopodiaceae, Cucurbitaceae, and Solanaceae. Of the 23 primer pairs constructed, 16, 22, and 19 produced amplicons from members of the Apiaceae, Brassicaceae, and Fabaceae, respectively. Twenty of 23 primer pairs were also functional in three monocot species of the Liliaceae and Poaceae. Successful amplification after extended-length PCR of 16 recombined ccSSR primer (RCCP) pairs suggested that the amplicons detected may represent complete coverage of the cucumber chloroplast (<italic>Cucumis sativus</italic> L.) genome. These analyses indicate that ccSSR and RCCP primer pairs could have utility for chloroplast genetic analysis, especially in plant species for which chloroplast genome sequence information is not available.

Fragment length variation of 20 ccSSR primer pairs and sequence substitution variation of four fragments (ccSSR-1, -7, -8, and -19) were used to investigate genetic relationships in Benincaseae (19 accessions of 9 genera) and other tribes (8 accessions of 6 genera) of the Cucurbitaceae family. Although convergence in fragment length occurs in more distant comparisons, a degree of positive congruence of taxa groupings based on bp-length and sequence substitution data was apparent. Moreover, these analyses indicate that distinct lineages exist within the Benincaseae tribe. Likewise, data support the hypothesis that the genus <italic>Benincasa</italic> is descended from an ancient African ancestor, and that the progenitor of the New World Sicyeae tribe shares a common ancestor with the <italic>Luffa</italic> lineage of the Old World Benincaseae.

Exact reciprocal F<sub>1</sub> and F<sub>2</sub> cross-progeny of Gy-14 x ‘Chipper’ and Gy-14 x ‘Little John’ matings were subjected to chilling treatments (4°C for 5.5 hrs at the first true-leaf stage). Significant differences between reciprocal progeny indicated that chilling tolerance is maternally inherited in cucumber. Sequenced polymorphic and paternally inherited random amplified polymorphism DNA (RAPD) bands AD2<sub> 1249</sub>, AV8<sub>916</sub>, and AV8<sub>969</sub> suggested that their origin is likely cucumber mitochondrial DNA, and that the chilling tolerance trait as identified is likely associated with the chloroplast genome, which is itself maternally transmitted in cucumber.
