Nitrous oxide (N2O) is a greenhouse gas produced in different microbial processes. Nitrous oxide reductases (N2ORs) are enzymes that catalyze N2O transformation to N2. Understanding the diversity of nitrous oxide reductases genes (nosZ ) is crucial to predict and manage emissions. This study developed oligonucleotides for amplifying a broader range of nosZ genes to assess their diversity in different environments. Sequences obtained were different from traditional nosZ sequences (62% amino acid similarity). Neither of the clone sequences fell into the traditional NosZ phylogenetic clades, but grouped with nontraditional NosZ sequences from Anaeromyxobacter spp., Magnetospirillum spp., Desulfitobacterium hafniense, and Dechloromonas aromatica. The detection of nosZ genes in the environment was achieved by ISRT-PCR/FISH using an internal fluorescently-labeled NosZ943 probe. We demonstrate the existence of many variants of nosZ gene. These variants could represent a high functional diversity for reducing N2O.
