Neuropeptides are important chemical messengers in the nervous system and can act as neurotransmitters, neuromodulators, or neurohormones. Decapod crustaceans are important model organisms for studying how peptide hormones regulate physiological processes. This project employed a unique sampling and detection methodology to study neurohormones in the crab, Cancer borealis. An in vivo microdialysis sampling technique was developed and coupled with capillary liquid chromatography mass spectrometry (MS) as well as matrix-assisted laser desorption ionization MS. This sampling and detection scheme resulted in the detection of many known neuropeptide masses in crab hemolymph, as well as the tandem MS sequencing of three known neuropeptides and one novel peptide. A comparison of microdialysis sampling with typical hemolymph preparation methods revealed more neuropeptides and fewer protein fragments in samples collected via microdialysis. This study demonstrates the ability of microdialysis to collect clean, neuropeptide-rich samples from the hemolymph of a live crab that can be detected using MS analysis. This method can easily be applied to detect neuropeptides or other low-mass analytes from other crustacean model organisms and complements current tissue-based techniques for sampling neuropeptides. As an in vivo sampling method, microdialysis with MS detection has the potential for real-time analysis of neuropeptides in a live, freely-moving organism.
