Systemic phaeohyphomycosis, known as "fluid belly", is one of the most important emergent diseases in sturgeon aquaculture. The etiologic agent is the saprobic, dematiaceous fungus Veronaea botryosa. Effective vaccines and chemotherapeutic treatments are currently unavailable, and currently, few diagnostic methods are available for this fungus. Moreover, the impacts of environmental conditions on fungal growth and conidium viability are presently unknown and the fungus is suspected to persist in the environment in the absence of fish hosts. To this end, specific quantitative PCR (qPCR) and reverse-transcriptase qPCR assays targeting the V. botryosa β-tubulin gene were developed to detect fungal DNA in tissues, and to investigate environmental persistence of V. botryosa in marine and freshwater microcosms at different temperatures. Additionally, the susceptibility of V. botryosa conidia to three disinfectants used in the aquaculture industry was determined, namely iodophors, chlorine and hydrogen peroxide. Probe specificity was confirmed by testing against common fish fungal pathogens and sturgeon tissue. The qPCR and rt-qPCR have limits of detection of 10 and 10000 conidia, respectively, which were not affected by the presence of non-target DNA from white sturgeon. Our results suggest V. botryosa remains viable in the environment without the fish host for >60 days and persisted in marine and freshwater microcosms maintained at 15˚C and 25˚C. However, the lower growth and persistence of V. botryosa in marine water were observed when compared to fresh water. In addition, disinfectants recommended for aquaculture are effective against V. botryosa and have application in biosecurity protocols for captive sturgeon. These methods can be used in future studies as non-lethal diagnostic methods and to quantify fungal loads in the environment.
