Arginine vasopressin (AVP) is a neuropeptide known to influence many rodent social behaviors. Experiment 1 assessed the effects on sexual behavior of AVP (1 ng) infusions into the olfactory bulbs of male rats, finding that AVP significantly inhibited the number of intromissions displayed compared to control animals. Experiment 2 sought to expand upon these findings by comparing groups receiving injections of AVP, a V1b vasopressin receptor antagonist, both AVP and the antagonist (1 ng in all cases), or vehicle. Additionally, immediately after testing, brain tissue samples were collected and were assayed for c-Fos expression in the medial amygdala. AVP alone and paired with the antagonist caused significant reductions in intromission. All drug-treated groups expressed significantly less c-Fos than controls. A model of olfactory processing is proposed whereby the antagonist depresses the function of mitral cells, the primary olfactory bulb efferents, while AVP activates granule cells, which inhibit mitral cells.
