Hormone-dependent pathways are critical in pregnancy, development, and reproduction throughout all stages of life. For this reason, when alterations are seen in pregnancy, development, and reproduction, hormonal changes are the first pathways investigated, specifically steroid hormones and peptide hormones. This research utilized a broad multi-tissue steroid hormone platform to better understand the importance and function of hormone-pathways in pregnancy, development, and reproduction in the presence of environmental, pharmacological, and nutritional influences. The boar and rat were selected as models for reproduction and maternal/fetal pregnancy outcomes, respectively. All tissue and fluid steroid hormone profiles were measured by ultra-performance liquid chromatography tandem mass spectrometry (UPLC/MS-MS). This is a novel, complex approach to steroid hormone profiling in animal models and the first report of a broad multi-tissue approach to steroid hormone profiling in boars and rats. Data generated demonstrate the existence of unique steroid hormone profiles within each tissue, and how in each organ they can respond differently and/or independently to environmental, pharmacological, and nutritional influences. The chapters contained herein: 1) expand the knowledge of tissue steroid hormone profiles, by exploring both free and conjugated steroids, and 2) examine each tissue as a unique entity that responds differently to nutritional, environmental, and pharmacological/toxicological influences.
