Brd2 is a double bromodomain-containing BET protein transcriptional coregulator functioning in multiple developmental pathways. Mammalian Brd2 is a proto-oncogene that regulates cell cycle, apoptosis, proliferation, and differentiation. In mice, Brd2 overexpression causes leukemias and lymphomas, but the role of Brd2 in blood development is rarely examined. I assessed impacts of brd2a knockdown on the transient wave of hematopoiesis of zebrafish within the Posterior Blood Island (PBI), in which erythrocytes and myelocytes are produced. Upon knockdown, mitosis decreases in the PBI at 24hpf, while apoptosis increases at both 24hpf and 30hpf, especially within erythromyeloid progenitors. Pronephric cells increase adjacent to the PBI, possibly due to misfating of common blood-kidney progenitors. The number of erythrocytes dramatically increases upon knockdown, implying a role for brd2a in erythropoiesis regulation. GO Analysis of RNA-seq confirmed that brd2a impacts expression of many blood-related genes. This study of brd2a expands our knowledge of its role in hematopoiesis.
