Mathematical modeling of tumor growth provides a theoretical framework for analyzing the dynamics and variability that play a role in tumor growth and treatment. The work in this thesis focuses on growth of metastases in a specific microenvironment, the lymphatic vessel. I first designed a mathematical model to understand the factors in this environment that will influence metastatic growth, and to determine the growth kinetics that ultimately result from these influences. I then examined the efficacy of treatment in this environment, to see what aspects of the lymphatic environment could alter the pharmacokinetics and treatment efficacy of a chemotherapeutic drug administered to tumor and metastases. These studies demonstrate the importance of lymphatic vessel architecture, lymph flow rate, and diffusion kinetics in lymph for the growth and treatment of intralymphatic metastases. The specific role of these components is discussed in detail in the thesis.
