The fields of neuropsychology, neuroimaging, and cognitive neuroscience have improved the understanding of brain functioning in complex cognitive tasks: however, little is known about the neurochemical underpinnings of complex cognitive ability. Proton magnetic resonance spectroscopy (1H-MRS) offers a unique non-invasive approach to the measurement of N-acetylaspartate (NAA), a putative markers of neuronal integrity. Previous studies have demonstrated that NAA predicts cognitive impairment in diseased subjects, although its relationship to cognitive functioning in healthy people is poorly understood. We hypothesized that NAA measured within frontal and occipito-parietal white matter would predict intellectual and neuropsychological functioning in a cohort of normal individuals.

We studied 49 subjects free from neurological, psychiatric, developmental or substance abuse disorders. Either STimulated Echo Acquisition Method (STEAM) or Point REsolved Spin echo Sequence (PRESS) was employed to sample three (12.6 cm3) voxel locations within left occipito-parietal and bilateral frontal white matter, utilizing a General Electric 1.5 Tesla clinical MR scanner. Standard measures of intelligence (Wechsler Adult Intelligence Scale-3) and neuropsychological performance were administered within 1 week of scanning. We found that NAA acquired within the occipito-parietal white matter was significantly related to Full Scale Intelligence Quotient (FSIQ) utilizing either STEAM [F(1,47) = 4.061, p < .05] or PRESS techniques [F(1,23) = 7.00, p < .02]. Similarly. we found that NAA acquired within the occipito-parietal white matter was significantly related to overall neuropsychological performance (Total-z score) utilizing either STEAM [F(1,47) = 17.34, p = .00011 or PRESS techniques [F(1,23) = 21.22, p = .0001].

The current finding of a strong association between NAA and cognitive ability in normal subjects suggests that 1H-MRS may be an important and sensitive tool for understanding normal brain functioning. Indeed, the relationship of NAA to broad based intellectual and neuropsychological ability appears to suggest that integration of numerous activation sites throughout the cerebral cortex---particularly frontal with posterior cortices---depend critically upon white matter interconnections sampled by our experimental voxel. Future research of both younger and older cohorts under a similar paradigm would help to establish the relationships between myelination, speed of information processing, and cognitive performance across the lifespan.
