Palatable foods in our environment activate the reward centres in our brain and tend to foster eating beyond caloric needs. Psychomotor stimulant drugs also regulate the reward pathway via dopamine (DA) release, typically causing appetite suppression. However, research shows that this response varies across individuals. ADHD is a disorder characterized by inattentiveness and distractibility, and is associated with low DA levels in the brain. Therapeutic treatment with methylphenidate (MP) increases DA levels to improve attention, but again, response varies. This study investigated whether genetic variation of three single nucleotide polymorphisms of the Casein Kinase 1 Epsilon Gene (Csnk1e) - a gene that regulates a protein found to affect drug sensitivity - results in changes in mood and eating variables in response to the stimulant drug, MP. By employing a drug vs. placebo, double-blind, cross-over design with 134 healthy adults, results indicated that individuals with C/C and C/G genotypes of the rs135745 SNP showed significant decreases in appetite ratings and food cravings in response to a MP induced snack-food challenge. Implications are discussed regarding proper drug treatment for ADHD to reduce side effects such as loss of appetite.
