Ixodes scapularis Say (Acari: Ixodidae) is the primary vector of Lyme disease which is now the most common arthropod-borne disease in the United States. Three new cell lines, IDE8 and IDE12 from embryos of northern specimens and ISE18 from southern specimens of I. scapularis were compared cytogenetically via conventional karyotyping, C-and G-banding, and nucleolar organizing regions (NORs). The standard karyotype in the three cell lines consisted of 28 chromosomes with 26 autosomes and XX (female) or XY (male) sex chromosomes. Constitutive heterochromatin (C-bands) was almost entirely restricted to the centromeric region. An additional interstitial C-band in chromosome 7 was an important characteristic of the three cell lines. The three cell lines had identical G-banding patterns. Interstitial NORs were identified in chromosomes 7, 10, and the X chromosome in the three lines. These findings support the notion that I. scapularis and I. dammini Spielman et al. are conspecific.Subjects--Topical Terms:

Melanoplus sanguinipes (Fabricius) (Orthoptera: Acrididae) is widely distributed throughout North America and is an economically important crop pest. Three new cell lines from the grasshopper M. sanguinipes were analyzed with respect to karyotypes, C-and G-banding, and NORs. In lines MSE4 and MSE8, the normal female cell karyotype was 2n = 24 (XX), and male 2n = 23 (XO). MSE3 cells were 2n = 22 (XX), female and 2n = 21 (XO), male. The chromosomes of the three cell lines had paracentromeric C-bands, and G-banding showed a stable and distinctive pattern. Four expressed NORs were interstitially located on chromosomes 2 and 4. Triploid, tetraploid, hypotetraploid, chromosome centric fusion, chromosome breakage and supernumerary B chromosomes were observed in the lines.

Indirect immunofluorescence assay of four tick cell lines, ISE5 and ISE6 isolated from southern populations of I. scapularis, IDE8 from northern populations of I. scapularis and RAE25 from Rhipicephalus appendiculatus (Neumann), were cross-reactive. However, the lines were distinguished from two insect cell lines. Three major proteins around 14 kDa, 22 kDa and 31 kDa separated with SDS-PAGE in the four tick cell lines. Multiple antigens between 12 kDa and 90 kDa were recognized by Western blot in the four tick cell lines. Some of the antigens were specific in the lines.
