A variable amount of commercially obtained Artemia cysts refuse to hatch as long as they are deprived from light. Upon a transient illumination of these cysts development is resumed. We investigated the mechanism underlying the light activation of these cysts. The complete sequence of events is not yet unravelled, but parts of the reaction chain are elucidated.Subjects--Topical Terms:

Concerning the photoreceptor, the results from two action spectra suggest the involvement of a porphyrin (probably a haempigment with abs.max. 450-525-575 nm).

The primary reaction in light activation in Artemia is a photochemical, oxygen consuming process. It seems that the photoreceptor needs to be oxidized to admit proper photo-activation. H

The involvement of signal molecules was investigated and the results indicate a possible regulatory role of cyclic nucleotides in light induced resumption of development. As well light-induced as H

induced resumption of development of light-deprived embryos of Artemia is accompanied by an immediate decrease in cAMP concentration. The concentration of cGMP however remains unaltered.

Next we demonstrated that light deprived cysts are not only developmentally but also metabolically arrested. This was shown by the absence of trehalose consumption, low respiratory rates and low ATP levels in light deprived cysts. Upon illumination of these cysts, we registrated an increase in respiration a decrease in trehalose- and an increase in ATP- and glycerol concentrations.

An electron microscopic study affirms the metabolic data since it reveals that light deprived cysts have round, electron dense mitochondria and no polyribosomes. Upon illumination, the mitochondria lose their circular outline and become elongated, and polyribosomes are formed.

A possible sequences of the demonstrated light induced events is proposed and the role of light as a trigger in relation to diapause and quiescence of Artemia cysts is discussed.
