Dietary cholesterol has been the subject of much recent study by lipid biochemists and others. However, the mechanism of removal of dietary cholesterol by-products by the liver plasma remains unknown. This dissertation is about results and analyses of experiments on the rate of hepatic uptake of cholesterol by-products in mice. The data consist of sixty-six continuous flow curves, approximated by discrete observations of twenty points each. Three approaches to the analysis of these data are presented. The first is an empirical non-parametric exploration using functional data analysis. The second approach is by way of competing parametric models: a compartmental model, which describes the mechanism of the biochemistry, and a catalytic model, which summarizes characteristics of the data. In the third approach, posterior densities of kinetic parameters of Bayesian hierarchical models are computed with Markov Chain Monte Carlo techniques. In each of the approaches, differential equations play an important role in the analyses. The computational techniques allow for investigation of relations among experimental covariates and key parameters of the models.Subjects--Topical Terms:
