Ascorbic acid (AsA) metabolism during iron-induced stress has been studied in non-synthesizing species. Few studies have examined responses in synthesizing species; fewer yet have measured effects on AsA biosynthesis. Tissue (AsA), hepatic peroxide and iron concentrations, and activity of L -gulonolactone oxidase (GLO; EC 1.1.3.8) were measured. Iron overload depleted plasma and hepatic AsA pools with concomitant up regulation of GLO activity in chicks fed 2000 mg/kg Fe for at least 4 weeks. Hepatic lipid peroxide concentrations increased with increasing iron overload. Treatment effects on dehydroascorbate reductase (EC 1.8.5.1) activity were not detected. A down regulation in GLO activity and a depression in plasma and tissue AsA were observed in chicks receiving low dietary antioxidants and excess iron. The relationship between dietary VE and AsA metabolism was examined in male Japanese quail fed a purified diet devoid of VE. Gulonolactone oxidase activity was up regulated with the addition of supplemental VE. The results showed that iron overload in chicks depleted tissue AsA and enhanced biosynthesis to compensate for increased oxidative stress. In contrast, iron overload reduced GLO activity in chicks fed a practical diet without supplemental VE and selenium. There was no evidence of up regulation of GLO activity in quail in the absence of dietary VE.
