Soil applied pesticides often lose their ability to control target pests following repeated annual applications to the same location. This loss of efficacy has been attributed to the rapid increase in the rate of pesticide degradation by microorganisms in treated soils compared to soils having no previous exposure to the pesticide or to other pesticides similar in structure. In soil, the herbicide EPTC (s-ethyldipropylcarbamothioate) is known to be biodegraded at enhanced rates. Several strains capable of using EPTC as a sole carbon source were isolated from soil by enrichment techniques and identified as members of the gram positive genus Rhodococcus. To facilitate plasmid DNA extraction, a procedure was developed to produce spheroplasts in Rhodococcus spp. and other gram positive bacteria utilizing the bacteriolytic enzymes mutanolysin and lysozyme.

The EPTC degrading Rhodococcus strains contained five plasmids: pNAS108, pNAS87, pNAS63, pNAS28, and pNAS9.1 (sizes 108, 87, 63, 28, and 9.1 kb, respectively). Curing experiments and electrophoresis analysis of the strains' plasmid content indicated that EPTC degradation is linked to the presence of pNAS108. This plasmid is conjugative and it can transform Rhodococcus strains lacking the ability to degrade EPTC to EPTC degraders. It can also integrate into the chromosome to generate an HFR strain capable of transmitting the chromosome incorporated gene by conjugation.

DNA hybridization indicated homology between the plasmids associated with EPTC degradation in all degrading strains and with chromosomal DNA sequences in strains that degrade EPTC while missing pNAS108. This observation confirmed the integration of the EPTC degradation gene(s) present on pNAS108 into the chromosome to form HFR strains.
