The association between Helicobacter pylori (H. pylori) and gastric disease---gastritis, ulceration and carcinoma---has been recognized for some time. Because the gastrointestinal tract may be viewed as extracorporeal, a method to target delivery of an active antibiotic agent could represent a significant advance in H. pylori eradication and disease treatment. The goal of this study was to develop a model system that would allow for the specific targeting of H. pylori through the use of antibody-conjugated polymer beads. The ability of such beads to interact with H. pylori was tested by creating a macromolecular, whole-cell ELISA system. Results were assayed using alkaline phosphatase-conjugated secondary antibody and p-nitrophenyl phosphate as a developing substrate. In addition, a solution phase assay, employing acridine-orange stained H. pylori cells was utilized to examine cell-bead interactions. Results demonstrate that polymer beads conjugated with Rabbit-anti-Helicobacter pylori polyclonal antibodies specifically interact with H. pylori. Conversely, polymer beads conjugated with non-specific Rabbit IgG show a significantly reduced interaction (Student t-test p < 0.05).
