The isoleucine-valine biosynthetic gene cluster of Salmonella typhimurium was characterized. Deletion analysis employing the transposable element Tn10 unambiguously showed the gene order to be ilvG-E-D-A-C. The ilvY gene, which encodes a positive effector of ilvC expression, was shown by Tn10 generated mutations to lie between ilvA and ilvC. Studies with F'ilv('+) plasmids demonstrated that the ilvB gene is not part of the ilvGEDAYC gene cluster.Subjects--Topical Terms:

The polarigenic property of Tn10 was used to define the direction of transcription of the ilvGEDA. The incomplete polarity of certain Tn10 insertion mutations was shown to result from transcriptional activity of the transposon itself rather than putative promoters within the ilvGEDA operon. The productive use of Tn10-derived transcripts of contiguous genes in the ilvGEDA operon was correlated with the presence of effective ribosome binding sites.

The parallel regulation of ilvGEDA from multicopy recombinant plasmids was demonstrated, and the gene products were shown to correspond in ratio to those produced from the chromosome. Expression of the individual genes is discoordinate and corresponds to a relative translational frequency for ilvG-E-D-A of 1:7:2:1.5. The basis of this discoordinate expression, particularly between ilvG and ilvE is most easily explained by qualitative differences between the ribosome binding sites associated with ilvG, ilvE and ilvD. The translation of the ilvA gene was shown to be strongly dependent upon translation of ilvD suggesting that there is translational coupling between ilvA and ilvD. The ilvA gene lacks a ribosome binding site, or alternatively, its ribosome binding site is masked by mRNA secondary structure which is disrupted by ribosomes translating the upstream ilvD sequence.

Two other transcriptional units were demonstrated in the ilv gene cluster, ilvY and ilvC. These genes were shown to be transcribed in the same direction as ilvGEDA. Expression of ilvY is apparently constitutive. The target site for the ilvY encoded positive regulatory protein is the ilvC promoter, which was shown to be immediately continguous with the 3'-end of the ilvY structural gene sequence. A cis-trans test utilizing the S. typhimurium ilvY mutant and various ilv-galK recombinant plasmids failed to demonstrate a cis-preference for the ilvY gene product.
