Natural antimicrobial peptides have potential application as food preservatives because these peptides destroy microbial cells by rapidly disrupting cell membranes, however, continued research is required to determine their appropriateness for food applications. The objective of this research is to evaluate the suitability of pleurocidin, an antimicrobial peptide originally isolated from the winter flounder (Pleuronectes americanus), as a potential food preservative and to utilize the Pichia pastoris system, for its recombinant expression.

This research was conducted in three phases. In phase 1, an antimicrobial spectrum and the cytotoxic activity of pleurocidin was determined by studying its efficacy against various foodborne pathogens and evaluating its effect on human cells. Pleurocidin was effective against Gram positive and Gram negative foodborne pathogens including; bacteria, yeasts and molds, without deleterious effects on human cells in vitro.

Phase 2 involved investigating the effect of intrinsic and extrinsic factors associated with food, on the efficacy of pleurocidin, using Vibrio parahemolyticus as test organism. Pleurocidin retained activity after autoclaving for 1 h (121°C, 15 psi), in high salt concentration (7.5% NaCl) and across pH range 4--8.

The efficacy of pleurocidin in food samples was investigated in phase 3. Based on the unique characteristics of salt and acid tolerance of pleurocidin, Escherichia coli O157:H7 and Vibrio parahemolyticus were used as target foodborne pathogens in apple cider and fresh shrimp respectively. A 2 log reduction of E. coli O157:H7 was shown in the presence of pleurocidin within three days (4°C) and complete inhibition within 15 days. Pleurocidin was also effective in reducing the growth of Vibrio parahemolyticus on shrimp.

Concurrently, the Pichia pastoris system was utilized for the recombinant expression of the pleurocidin gene. Polymerase chain reaction, DNA sequencing, and reverse-transcriptase reactions showed that the pleurocidin coding sequence was successfully integrated into the Pichia pastoris genome and was transcribed by both the wild type and slow methanol utilizing strains of Pichia pastoris. However, the level of pleurocidin expressed was below detection levels.
