Kaposi's Sarcoma associated herpesvirus (KSHV) establishes latent infection in dividing cells. To persist, viral episomes must replicate and properly segregate to progeny nuclei. Latency-associated nuclear antigen (LANA) mediates viral persistence by tethering episomes to chromosomes during mitosis. LANA contains two independent chromosome association regions located in the protein's N- and C-terminal regions. N-terminal LANA chromosome binding is necessary for both episome replication and persistence. However, little is known about C-terminal LANA chromosome binding. In this dissertation, we have used mutational analysis to characterize the C-terminal LANA chromosome binding region and investigate its role in episome persistence. We demonstrate that C-terminal LANA concentrates to paired dots at pericentromeric and peri-telomeric regions of a subset of mitotic chromosomes through residues 996-1139. A deletion abolishing specific DNA binding did not affect chromosome targeting, indicating that LANA's localization does not occur by direct binding to chromosomal DNA. Instead, C-terminal LANA chromosome binding likely occurs by interaction with a cell factor(s) that concentrates at pericentromeric and peri-telomeric chromosome regions.
