Epidemiological evidence clearly indicates that there are health benefits associated with the consumption of fruits and vegetables. Although numerous in vitro studies demonstrate their antioxidant capabilities, it remains unclear if these activities translate to physiological mechanisms resulting in the health promoting effects. What is clear is that flavonoids undergo extensive metabolism and that the biological activity from these compounds is likely due to metabolites. Understanding the disposition of flavonoids in the body is critical for assessing the role of fruits and vegetables in public health and disease prevention.

In plants, the range, levels and types of flavonoids vary greatly between varieties of fruits and vegetables. The following dissertation describes two manuscripts pertaining to the characterization of flavan-3-ols (i.e. the procyanidins) in canned Ross clingstone peaches and dried Deglet Noor dates. Procyandin monomers through decamers were characterized in peaches using LC/ESI-MS/MS methods. Results from these studies indicate that the procyanidins in peaches are considerably reduced during thermal processing (i.e. canning) by the migration of these compounds into the canning syrup. Analysis of Deglet Noor dates indicates that this palm date fruit is a rich source of procyanidins and has the sulfated forms of the flavonoids.

With the recognition of the extensive metabolic transformation of flavonoids in vivo and the importance of their characterization, we developed LC/ESI-MS/MS methodology to profile the range of flavonoid metabolites in human urine taken after the onion consumption. Twenty one quercetin-related metabolites were identified in this study with the inter-individual variation in urinary quercetin metabolic profiles, suggesting that metabolite profiles can be used to explore individual biotransformation capacity.

In related studies, the abilities of mice and humans to form glutathione-related metabolites of quercetin were investigated. In humans, several mercapturic acids were identified in urine as derivatives of colonic microbial degradation products of quercetin, proposing that quercetin is likely glutathionylated, secreted in bile where it becomes a substrate for colonic microflora and subsequently reabsorbed and circulated in plasma. This finding points to the possibility that microbial degradation products may also have biological roles and warrants new investigations in this area.
