A significant discrepancy exists among currently employed methods of enumeration of toxic microalgae. Species of the genus Alexandrium (Hahm) Balech, which are well known producers of PSP toxins in coastal waters, are extremely difficult to distinguish reliably under conventional microscopy. The application of species-specific molecular probes has proven to be a unique solution to accurately identify these and other toxin-producing species. To determine their effectiveness, two rRNA-targeted probes were applied to species of Alexandrium grown in culture and to field samples from coastal regions of Nova Scotia, Canada. This method was compared with three other light-microscope methods and discrepancies were found to occur among them when applied to the same sample. To elucidate reasons for the discrepancies, experiments were conducted with cultured Alexandrium tamarense and A. ostenfeldii. Possible causes of this phenomenon have been speculated to be the type of fixation method used and physiological status of the target cell. (Abstract shortened by UMI.)
