Naive CD4+ T cell-derived, IL-2-secreting primed precursor CD4 T (Thpp) cells represent an expanded pool of uncommitted T cells in vivo in both human and mouse. To discover additional functions of these cells and to identify specific surface markers, Affymetrix Genechips were used to identify genes uniquely expressed in Thpp cells, particularly after induction.

We compared gene expression of induced and uninduced Thpp cells using naive, Th1 and Th2 cells as controls. Except for IL-2 and Lymphotoxin, activated Thpp cells do not express regular cytokine genes (IFNgamma, IL-3, IL-4, IL-5, IL-6, IL-9, IL-10, IL-13, GM-CSF), but they do express high levels of several chemokine genes (TCA3, MIP-1alpha, MIP-1beta, MIP-1gamma, RANTES and Lymphotactin). This was confirmed by protein secretion assays and chemokine bioassays. However, activated Thpp cells did not directly influence the polarization of other CD4 T cells. Therefore activated Thpp cells may recruit immune cells without biasing differentiation or effector functions of other CD4 T cells.

Chip analysis data also predicted that a GPI-linked protein, Neuritin, might be Thpp-specific. PCR assays confirmed that Thpp cells do express the neuritin gene, however, the expression was neither Thpp-specific nor induction-specific.

Both the chip data and FACS analysis indicated that in vitro mouse Thpp cells expressed higher levels of CD73 than Th1 or Th2 cells, but lower levels of Ly-6A/E. CD73 is an ectonucleotidase that produces the anti-inflammatory mediator adenosine if extracellular AMP is present. CD73 and Ly-6A/E also identified in vivo Thpp-like cells in C57Bl/6 mice, as CD73+Ly-6A/E- memory CD4 T cells isolated directly ex vivo produced IL-2 and MIP-1alpha, but much less IL-4 and IFNgamma in response to antigen. These cells were enriched in lymph nodes, and showed flexibility in further differentiation. Our study has thus identified new markers to define Thpp-like cells in mice of the Ly-6.2 haplotype.

Taken together, our data show that Thpp cells produce several potent chemokines that may attract other immune cells without strongly regulating effector functions. Selective expression of CD73 may dampen local effector functions, and in conjunction with Ly-6A/E may be invaluable for analyzing the contribution of Thpp during infection and vaccination.
