Techniques were developed to collect and prepare microbial samples from intestinal digesta and mucosal surfaces from early-weaned piglets. The washing procedure was developed for preparation of bacteria attached to the mucosal surface. A new commercial kit (QIAampRTM, QIAGEN) was proved to be effective for DNA extraction from intestinal microflora in pigs. A polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) method demonstrated a high level of reproducibility and sensitivity for characterization of pig intestinal microflora. Effects of including antibiotics, two acid mixtures and one herbal extract product in the diet on growth performance and intestinal microflora diversity were investigated using piglets weaned at 16 days of age. Supplementation with acid mixture-2 (containing 1% lactic acid) increased average daily gain (ADG) as compared to the non-medicated control (P < 0.05) and brought it to a level similar to the medicated control (P > 0.05) at week 2 post-weaning. Supplementation with these feed additives inhibited a range of intestinal bacteria. In addition, the ileal microflora exhibited a lower diversity than those in the cecum and colon. Diversity of the intestinal microflora increased from weaning to day 14 post-weaning.
