The goal of these studies was to determine the role of the D1-dopaminergic receptor system of the suprachiasmatic nucleus (SCN) in maternal/fetal entrainment. In the first part of the dissertation, the ontogenic anatomy of the D1 system was studied. A receptor-transmitter mismatch was found between the prenatal appearance of D1-dopamine receptors in the SCN and the postnatal innervation of the SCN by tyrosine hydroxylase-positive neurons, suggesting a paracrine action of dopamine in the fetal SCN. Using a transplantation paradigm, we asked whether a D1-dopamine receptor agonist, SKF 38393, could set the phase of the grafted circadian pacemaker. The results demonstrated that SKF 38393 unambiguously set the phase of restored locomotor rhythms in adult hamsters bearing fetal whole tissue grafts containing the SCN, supporting the hypothesis that activation of the fetal dopaminergic system in the SCN plays an important role in the mechanism of maternal/fetal entrainment. And finally, the long-term effects of perinatal cocaine exposure on the adult circadian timing system were tested. Cocaine-treated animals showed altered phase responsiveness to light pulses given at specific times during the circadian cycle compared with saline control animals. The ability of early cocaine exposure to produce long-lasting effects on the phase shifting capabilities of the adult clock could have significant clinical implications.
