Preliminary studies of DNA sequence variation in populations of Drosophila ananassae from Australia and Samoa have shown evidence for high levels of population structure (Schug, personal comm.). The patterns of isolation found in populations throughout the geographic range in Southeast Asia and the South Pacific Islands (Tobari, 1993) may reflect adaptation to local geographic regions (Chen et al., 2000; Stephan et al., 1998). This may be accompanied by or preceded by pre-mating reproductive isolation, which is reflective of divergence in mating behaviors and is believed to be a mechanism of speciation.

Multiple choice experiments and a discrimination index were used to quantify the levels of sexual isolation among populations of D. ananassae from Indonesia, Australia and Samoa. The results of the study show strong pre-mating reproductive isolation exists among Samoan and Australian populations. Complete pre-mating reproductive isolation was observed between the light and dark populations on Samoa. The sexual isolation found between the two populations supports the historical proposal that reinforcement may be occurring on Samoa. The pre-mating reproductive isolation observed between the light form on Samoa and the northern Australia population also reinforces Futch's (1966) hypothesis that the lighter flies from these two locations may have shared a common ancestry different from the other D. ananassae populations in this region.
