The effects of 1,25(OH)2D3, PTH and 25(OH)D 3 on phosphate or calcium uptake were studied in cultured chick enterocytes using a period of 10 min after hormone addition. Cells treated with 130 pM 1,25(OH)2D3 showed an increase in 32P uptake. Similar studies with 65 pM bPTH(1-34) or 100nm 25(OH)D 3 resulted in increased 45Ca uptake. Confocal microscopy using the endocytotic marker dye, FM 1-43, revealed that membrane trafficking is rapid enough to contribute to ion transport.

Studies were undertaken using the PKC and PKA inhibitors, chelerythrine and Rp-cAMP, respectively. Results with 1,25(OH)2D3 and these inhibitors were complex, whereas preincubation of enterocytes with Ab099 (against a membrane receptor for the seco-steroid) abolished 1,25(OH) 2D3-stimulated 32P uptake, and FM 1-43 redistribution. The stimulatory actions of bPTH(1-34) or 25(OH)D3 on 45 Ca uptake and membrane trafficking were decreased by Rp-cAMP.
