Scope and method of study. Research studies have indicated that there is a relationship between chronic inflammation and prostate cancer progression. Pro-inflammatory mediators such as interleukin-8 (IL-8), interleukin-6 (IL-6), and vascular endothelin growth factor (VEGF) have been proposed to play a role in the development of hormone resistant prostate cancer in part because they promote angiogenesis. The growth of new blood vessels is crucial for the pathogenesis of prostate cancer and other forms of cancers, because without them, nutrients needed for cell growth and proliferation will be unavailable. Previously, our laboratory has shown that white button mushroom extracts inhibited IL-8 secretion by THP-1 cells.

The aim of the current study was to investigate the effect of extracts from White button mushrooms (WBM), Shiitake mushrooms (SM), and Portabella mushrooms (PM), prepared by ethanol precipitation, on the secretion of IL-8, IL-6 and VEGF and the proliferation of PC3 and LNCaP cancer cell lines. Cells, were incubated without and with mushroom extracts, for 3, 6, or 24 h in various culture mediums. Various cytokines were measured in the supernatants of mushroom extract-treated and untreated cultures by ELISA. NF-kappaB assays were conducted to examine the potential mechanism of inhibition. Cell viability assessed by trypan blue exclusion test and cell proliferation measured by tetrazolium bromide reduction (MTT) were also performed.

Findings and conclusions. We reported that mushroom extracts inhibited the secretion of pro-inflammatory cytokines by PC3 cells in a dose dependent fashion. IL-8 and IL-6 were undetectable in LNCap cells. Mushroom extracts also reduced VEGF secretion by LNCaP cells. Mushroom extracts inhibited the proliferation of PC3 and LNCaP cells. Preliminary data suggest that WBM and SM, but not PM extracts may affect cell proliferation and cytokine secretion through NFkappaB activation. Taken together, our data suggest that mushroom extracts modulate prostate cancer cell growth and function. More experiments are planned to investigate the mechanisms by which mushroom extracts inhibit cytokine secretion by PC3 and LNCaP cells and if these results are reproducible in vivo and what these findings imply in regards to mushroom consumptions and prostate cancer pathogenesis.
