As part of the broader objective to improve almond production, this study was initiated in order to investigate the potential of selected almond scion and rootstock cultivars in the transformation for disease resistance and to develop a protocol for efficient regeneration of transformed cells based on the established walnut somatic embryo gene transfer system. Much of the work was on the improvement of somatic embryogenesis to obtain highly embryogenic lines that could be used for transformation. The immature (68-88 d post-anthesis) cotyledon explants exhibited various morphogenic responses to different media. The frequency of somatic embryogenesis was genotype- and age-dependent. Some treatments were effective in inducing somatic embryogenesis up to the cotyledonary stage, but certain stages of somatic embryos seemed to require a different set of treatments to promote normal differentiation and germination. Transfer to hormone-free medium allowed repetitive embryogenesis and consistent production of secondary somatic embryos was observed from a clonal PA hybrid OP. This hybrid rootstock could be an excellent cultivar for transformation due to its high capacity for regeneration and good horticultural characteristics. Improvement of this rootstock by incorporating genes for disease resistance can have broad applications to almond production. Future studies should consider improving the regeneration frequency and somatic embryo conversion in this hybrid rootstock to increase chances of successfully transforming and recovering transgenic plants.
