Investigating the presence of bovine leukemia virus (BLV) in the colostrum of cattle helps determine mechanisms of transmission of the virus to calves. Colostral cells were separated through two density gradients, cultured to induce BLV expression and immunostained to determine immune cell types or the presence of the viral capsid (CA) protein. DNA extracted from colostral cells was analyzed to determine the total number of cell genomes and the number of copies of the BLV pol gene encoding the reverse transcriptase. Systematic evaluation of reagents and staining conditions allowed specific staining of colostral cells in stored samples. Background staining was reduced using different blockers (fish protein, bovine serum albumin, and nonfat dried milk). Antigen retrieval by heating in buffers of pH 3, 6, and 9 was investigated when necessary. Immunostaining was reported by an enzymatic color reaction. Cells were counted that were positive for the viral CA protein or one of the following cell proteins: CD3, CD4, CD5, CD8 and WC1 (T cell markers); CD21 (B cell marker); or CD14, CD172a, and MHCII (myeloid cell markers). The denser cell fractions contained mostly lymphocytes while the less dense fractions were enriched with myeloid cells. Although monocytes were mostly found in the least dense fractions, a few monocytes were present in the denser fractions. Real-time analysis of DNA by the polymerase chain reaction revealed that the BLV provirus was present in all colostral cell fractions from 12 of 20 BLV-seropositive cows and was present in one or more cell fractions from 4 additional animals. Cells having densities of 1.04-1.05 g/cc had 1 copy of BLV DNA present for every CA+ cell detected. Denser cells (1.05-1.06 and 1.06-1.08 g/cc) contained 3 and 6 copies of BLV DNA per CA+ cell, suggesting that each infected cell contains more than one copy of BLV DNA or that fewer cells express the CA protein in culture than the number containing the BLV provirus. Learning how immune cells carry BLV in colostrum will help develop ways such as pasteurization to prevent virus transmission to neonatal calves during colostrum ingestion.
