Thesis (Ph.D.)--Cornell University, 2007.
The overarching aspiration of this dissertation is the manipulation and utilization of macromolecules through cross-linking. In the first example, creation of a nontoxic, biodegradable coating using calcium alginate and FD&C approved dyes is described. This non-covalently cross-linked polymeric coating is robust but is rapidly removed upon treatment with disodium ethylenediamine tetraacetate (EDTA). The coating can be used as an easily removable and environmentally friendly logotype on an artificial turf surface. In the second example, formation of microencapsulated, site-isolated polymeric catalysts by covalent cross-linking of a macromolecule at the interface of oil-in-oil emulsion is described. A polar polymer, polyethyleneimine, is dissolved in polar solvent and dispersed in non-polar solvent. Upon emulsification, 2,4-tolylene diisocyanate is added to cross-link polyethyleneimine at the interface of the oil-in-oil emulsion droplet. The obtained microcapsules with polyurea shells and containing unreacted polyethyleneimine can be used as a site-isolated catalyst. An important intermediate in the synthesis of gamma-amino acid analogs can be obtained if this site-isolated catalyst is used in tandem with a nickel-based catalyst. Interestingly, the tandem catalysis would not have worked if the polyamine catalyst had not been encapsulated; the amines would have complexed nickel and poisoned the catalyst. The tandem catalysis described calls for the use of two miscible solvents; methanol and toluene. The polymer is not soluble to the same degree in both of these solvents and because of that a phase separation occurs inside of the capsules. This phase separation abides by the well-known Flory-Huggins theory and is general. With the proper choice of polymer and solvents, many different microenvironments can be produced this way. New artificial microenvironments like these could impact areas from chemical synthesis to drug delivery.
