This dissertation represents an effort to characterize immunostimulatory (monocyte/macrophage enhancing) compounds from the following dietary supplements and natural products: food-grade microalgae, Aloe barbadensis, Astragalus species, Echinacea species, Podophyllum species, and marine/aquatic algae. A transcription-factor based reporter gene assay measuring activation of nuclear factor kappa B (NF-kappa B) in THP-1 cells was used to evaluate monocyte activation. A secondary bioassay detecting induction of mRNAs for interleukin-1 beta and tumor necrosis factor alpha was used to confirm immunostimulatory activity.

An aqueous extract from food-grade Aphanizomenon flos-aquae microalgae contained monocyte-activating properties that were transduced through pathways similar to LPS-dependent activation. The compound responsible for this activity is a new high molecular weight polysaccharide, called Immunon, that can be recovered with a 5% yield and an EC50 value for NF-kappa B activation of 20ng/mL. Immunostimulatory polysaccharide preparations were also isolated from two other food-grade microalgaes, Spirulina platensis and Chlorella pyrenoidosa.

Immunostimulatory properties of Aloe vera juice were compared with acemannan, a carbohydrate component from aloe purported to have immune enhancing effects. Results demonstrated that acemannan is a weak activator of monocytes and that its low activity is due to the presence of a new, potent polysaccharide named Aloeride which is present in trace amounts in acemannan preparations.

Crude non-polar extracts from Echinacea species were inactive for monocyte activation. However, fractionation resulted in two regions of activity, immunosuppressive and immunostimulatory. The immunostimulatory fractions both enhanced LPS-induced macrophage activation and also acted as independent activators. From Astragalus species 19 cycloartane-type triterpene glycosides were analyzed and all were inactive except astragaloside I. Podophyllotoxin and specific analogs did not function as independent activators, but instead selectively enhanced or suppressed LPS-induced gene expression. Several hundred extracts of marine/aquatic algae were also evaluated for monocyte activation. Active extracts were fractionated to obtain purified substances with potent immunostimulatory activity (EC50 values of 10ng/mL).
