Fluorescence based sensors are of extreme importance and have found applications in various fields. While the demand for useful fluorescence sensors is acute, the conventional target oriented approach has limited the scope and rapid growth of novel sensor discovery. To make a breakthrough in this field, we introduced a new diversity oriented fluorescence library approach (DOFLA) using combinatorial fluorescent compound libraries with huge structural diversity.

Three fluorescent dye libraries with high purity were prepared by solid phase synthesis, including qunaldinium, benzimidazolium and anillno-indolium scaffolds. These compounds were subjected directly to fluorescence based high-throughput screenings with diverse analytes.

Two specific and unique fluorescence sensors for the enzyme alpha-chymotrypsin were discovered, which showed distinctive responses only to chymotrypsin but not the other several common serine proteases or related protein including chymotrypsinogen, elastase, subtilisin and trypsin. These compounds were believed to help in the future study of chymotrypsin conformation changes.

The first highly selective fluorescence turn-on GTP sensor-GTP Green, was discovered through screening of the benzimidazolium dye library. This compound showed more than a 90 fold fluorescence increase only with GTP, while less than 2 fold fluorescence increase to other 16 related nucleosides and nucleotides were observed.

Two remarkable fluorescence sensors, Heparin Orange and Heparin Blue, were discovered for Unfractionated Heparin (UFH) and Low Molecular Weight Heparin (LMWH), which were clinically used as a major anticoagulant to prevent thrombosis and to treat thrombotic disease. Impressive fluorescence red shift and fluorescence turn-on were observed by the two compounds and the phenomena were so obvious that they could be easily distinguished by even naked eyes. These compounds have the potential to be applied to monitoring heparin levels in clinical plasma sample for point-of-care detection.

Taking all these discoveries together, specific and unique sensors for a broad range of analytes from polymers and proteins to signaling small molecules such as GTP could be discovered from the library pools, demonstrating the universal applicability of this approach.
