<italic>Para</italic>-aminophenol (PAP) nephrotoxicity is generally associated with necrosis, but previously we observed DNA fragmentation, characteristic of apoptosis, in kidneys from rats treated with PAP. The mode of cell death has important consequences in that necrotic cells elicit inflammatory responses, whereas apoptotic cells do not. We were interested in determining the extent to which apoptosis contributes to PAP-induced cytotoxicity. For many toxicants, both types of cell death can be observed with apoptosis predominating at low toxicant concentrations and necrosis predominating at higher toxicant concentrations. These studies employed LLC-PK<sub>1</sub> cells, a renal epithelial cell line from pig kidney and compared the effects of different concentrations of PAP on LLC-PK<sub>1</sub> cells. Changes in cell membrane integrity, nuclear morphology, and DNA integrity were assessed. The results showed that LLC-PK<sub>1</sub> cells treated with lower concentrations of PAP (0.05–0.1 mM) exhibit characteristics of apoptosis: apoptotic bodies, chromatin condensation, and DNA fragmentation. Higher concentration of PAP (0.3 mM) was primarily associated with necrosis, showing early loss of cell membrane integrity without nuclear changes. Both GSH and caspase inhibitors can attenuate PAP-induced chromatin condensation and nuclear fragmentation, suggesting that oxidative stress may be involved in PAP-induced apoptosis and caspases were activated in the process.
