Anthropogenic activity in the Amazon region of Brazil is an ever-increasing source of environmental stress. Mining; deforestation; and human development have all impacted Amazonian ecosystems to some extent, although studies have generally only focused on the effects of mercury contaminants arising from gold mining activities. This study examined the activity of the hepatic biomarker, glutathione S-transferase (GST) with regard to varying degrees of human and industrial activity along the Rio Negro, Amazonas, Brazil. Fish were collected from 15 sites on the Rio Negro and livers were removed, quick-frozen in liquid nitrogen and stored until analysis. GST activity in the post mitochondrial supernatant was determined toward 1-chloro-2,4-dinitrobenzene and this activity was examined for significant variation in fish species, sampling sites and sediment metal concentrations. The most significant factor affecting variation in fish GST activity was the fish species. Site and sediment metal load also modulated GST activity, but to a lesser degree.
