Average lifetime values tauAv1 and tauAv2 during rigor, contracting and relaxation states of phalloidin-FITC labeled myofibril samples were acquired by measuring the fluorescence decay function in a Time Correlated Single Photon Counting Spectrometer. The longest lifetime values occurred during rigor and the shortest during contraction, while intermediate lifetime values were the longest detected during relaxation. Thus, the lifetime values observed were highest during the periods of least motion of the molecular motors. The consistency observed in repeat data acquired provides confidence for its use for analysis of fluorescent mechanisms.
