This study has accomplished a 3D alignment system that handles large-scale confocal volume data and denotes the inner tube structure of tumor microcirculation vascular tissue. It performs 3D alignment and reconstruction of large-scale confocal volume in virtual environment and a computer cluster. The structure and workflow of the system are presented in details along with 3D alignment method of large-scale confocol datasets and the representation and highlight of the tube shape of VM pattern. The design and algorithms are described within each module of the client, the server and the network. The results of this study, including images and analysis of the three main characters of our system, are presented and compared with the similar functions of other confocal alignment systems.

First, for the character of 3D alignment of large-scale confocal volume data, the result images demonstrate that the system can reconstruct the full volume structure and keep the high-resolution display for the detail at the same time. Compared to other alignment system, our 3D reconstruction not only works as a tool to display the final result but also gets involved in the alignment process directly as the real time feedback. The time spent in each step of rendering is also analyzed.

Second, for the character of alignment in the specimen's original physical section, the snapshots during the alignment process are shown to reveal the match between a pair of sections in both full view and zoom-in view and the entire confocal volume display effect before and after alignment.
