The present dissertation is organized in three manuscripts. In the first manuscript, we compared the peptidase activities of the excretory/secretory antigens of oncospheres of T. solium and the related, but non-pathogenic, T. saginata. T. solium and T. saginata oncospheres were cultured, and the spent media of 24-, 48-, 72-, and 96-hr fractions were analyzed. Activities for serine peptidases, cysteine peptidases, and aminopeptidase B, were tested fluorometrically. All peptidases were present in both T. solium and T. saginata; however, different patterns of activity were found. A chymotrypsin-like peptidase showed the highest activity in both parasites.

The second manuscript describes a novel cathepsin L-like protein fraction, purified from T. solium cysticercus fluid, which contains a 53 and a 25 kDa protein with cathepsin L-like activity. A western blot based on our cathepsin L-like purified fraction was tested. The sensitivity of the assay was 96% for multiple cysts patients and 78% for single cyst patients. Specificity was 98%. Lectin blots on the purified cathepsin L-like antigens confirmed the presence of several carbohydrates. The cathepsin L-like purified fraction elicited an IgG antibody response in rabbits and pigs, but was not protective against porcine cysticercosis.

The third manuscript describes the standardization and evaluation of ELISA, based on the cathepsin L-like protein fraction. The overall sensitivity and specificity of the cathepsin L-ELENA was 95% and 92%, respectively (the sensitivity was 84% in patients with a single cyst and 98% in patients with more than 1 cyst), and there was about 8% cross-reaction against Hymenolepis nana and Enterobius vermicularis.

In conclusion, a protein fraction with cathepsin L-like activity, purified from the cyst fluid of T. solium, were useful antigens for the immunodiagnosis of human cysticercosis. Although this antigen elicited IgG antibodies in rabbits and pigs, it was not protective against a T. solium proglottids challenge in pigs.
