With advanced understanding of the recently discovered CRISPR (clustered regularly interspaced short palindromic repeats) /Cas (CRISPR-associated) gene editing system, we are now able to perform more precise editing on desired loci. Nevertheless, current plant genome editing methods often require a tissue culture approach, which usually takes a great amount of time and labor to achieve. In this study, we aimed at utilizing the CRISPR technology to modify the tomato genome, and develop a novel method to select for mutagenesis with counter selectable markers (CSMs), in which plants undergo the processes of mutagenesis, decapitation, and selection, resulting in regeneration of new shoots. The CSM is designed to lead to the regeneration of mutated plants only. We have conducted tests on several potential CSMs and established a feasible way to deliver selecting agents to plants. We have also performed multiplex editing by targeting two genes in tomato, and the result was promising.
