As the number of patients requiring valve replacement increases, there is an increasing need to develop tissue-engineered heart valves (TEHV) to overcome the limitations of current surgical treatments. Bioprinting is becoming a common technique for producing complex 3D scaffolds such as TEHVs. Common bioink hydrogels used for the construction of 3D scaffolds are brittle and cannot mimic the mechanical behavior of soft human tissues such as skin, skeletal muscle, and blood vessels. To address this, a triblock biodegradable polymer of polycaprolactone, polyethylene glycol, and diacrylate (PCL−PEG−PCL)-DA was developed. This hydrogel formulation has high elasticity and flexibility allowing for the transfer of mechanical waves through the scaffold. Many cell types such as valvular interstitial cells (VICs) in native heart valves require mechanical loading for proper phenotypic modulation. The elasticity of PEG-PCL-DA hydrogels will improve the conduction of mechanical stimulation to seeded VICs which would increase scaffold remodeling. As a proof of concept for the PEG-PCL-DA hydrogel formulation, we 3D print a tissue-engineered heart valve and construct a bioreactor to simulate in vitro mechanical loading.
