In crucifers, sporophytic control of pollen self-incompatibility (SI) phenotype and the occurrence of dominance/recessive interactions among S haplotypes in pollen were not understood at the molecular level. With the isolation of SCR (S-locus Cys-rich), the male determinant of SI, it became possible to test two major models of sporophytic control of SI that were proposed in the 1970s and late 1990s. In an attempt to test these models, I used two approaches. A transgenic approach, in which SCR was expressed in Brassica oleracea either sporophytically under the control of a tapetum-specific promoter or gametophytically under the control of a pollen-specific promoter, was inconclusive. However, another approach, in which Arabidopsis lyrata SCR alleles exhibiting inherent differences in expression pattern were analyzed by in situ hybridization, demonstrated that tapetal expression of SCR is sufficient to confer pollen SI specificity. This analysis also showed that, in heterozygotes, recessive SCR alleles are silenced in the presence of a dominant SCR allele, thus providing a molecular explanation for dominant/recessive interactions of S haplotypes in anthers. Successful fertilization in flowering plants is dependent on highly specific pollen-pistil interactions beginning at the stigma epidermis and culminating in fertilization of ovules in the ovary. Very little is known about the mechanism and regulation of pollen adhesion, pollen hydration, and pollen tube growth and guidance within the pistil. To identify genes having potential roles in these processes, I used a method that combines genetic ablation of cells of the stigma epidermis and transmitting tract with genome-wide transcript profiling. This analysis identified 115 and 34 genes predicted to be express specifically in stigma epidermis and in transmitting tract, respectively. The identity, validation, and initial characterization of these transcripts are presented.
