Homogenous gold catalysis has emerged as a powerful methodology for constructing benzo-fused heterocycles such as benzofurans, benzothiophenes, and indoles. These heterocycles are privileged molecular scaffolds in medicinal chemistry, with currently marketed FDA-approved drugs containing the benzothiophene core including Zileuton (asthma symptoms controller), Raloxifene (osteoporosis treatment), and Sertaconazole (antifungal). Examples of FDA-approved drugs containing the benzofuran core include Naloxone (opiate overdose treatment), Amiodarone (arrhythmia preventative), and Vilazodone (selective serotonin reuptake inhibitor). Although there is great interest in generating these scaffolds, synthetic approaches to these heterocycles rely predominantly on stoichiometric or sensitive reagents. Advances in the field of organometallic chemistry have shown that homogenous gold catalysis could be a superior route for the construction of these heterocycles from functionalized alkynes. Presented herein is a detailed discussion about the development and utility of gold(I) complexes for the synthesis of benzo-fused heterocycles with various degrees of substitution. A catalytic method for the synthesis of 2,3-disubstituted benzofurans from functionalized 2-alkynylaryl ethers has been developed. This approach utilizes air- and water-stable NHC (N-heterocyclic carbene) gold(I) complexes as catalytic agents. These transformations proceed smoothly with low catalyst loadings and mild reaction conditions and tolerate a diverse group of functional groups with various steric and electronic properties.
